PADA BEBERAPA IKAN BUDIDAYA

ABSTRAK

       Perkembangan budidaya ikan di indonesia mengalami hambatan dengan berjangkitnya wabah penyakit KHV (Koi Herpes Virus) sejak 2002 yang telah merugikan ratusan miliar rupiah dan masih terus berlangsung hingga saat ini. Tujuan penelitian ini adalah untuk mengetahui jenis-jenis ikan yang dapat terinfeksi tetapi dapat berpotensi sebagai pembawa penyakit KHV. Ikan yang diuji adalah ikan konsumsi dan ikan hias meliputi ikan mas (Cyprinus carpio), ikan mujair (Tilapia mosambica), ikan tawes (Puntius javanicus), bawal (Colossoma spp), ikan mas koki (Carassius auratus), dan komet (Carassius carpio),. Pengujian dilakukan dengan melihat gejala kelinis dan pemerikasaan Polymerase Chain Reaction (PCR). Selanjutnya ikan di aklimatisasi dan diinjeksi dengan virus KHV. Pengujian inang alternatif KHV masing-masing dilakukan 3 (tiga) kali ulangan dengan teknik uji kohabitasi (pemeliharaan bersama). Selama pengujian dilakuakan penurunan suhu 3-4^oC dan penaikan pH hingga 8. Sebelum diuji coba, ikan tidak menunjukan gejala klinis terserang penyakit dan dalam uji PCR tidak menunjukan terinfeksi KHV. Ikan mas pada uji pendahuluan menunjukan tingkat kematian 40% hingga 80%, masing-masing untuk isolat Padang dan Cirata Jawa Barat. Pada kohabitasi dengan ikan mas yang terinfeksi KHV ikan mas menunjukan tanda-tanda serangan KHV dan menimbulkan kematian sebesar 20% hingga hari ke 9 dan hingga 80% pada hari ke 10. Ikan koi dapat menjadi inang yang cocok bagi KHV. Inang alternatif lain (koki, komet, tawes, mujair, gurame, bawal) yang dikohabitasi dengan ikan mas terinfeksi KHV tidak menunjukkan gejala klinis terinfeksi KHV. Sehingga tidak dapat menjadi inang bagi KHV.

WITHDRAWAL TIME ~ OXYTETRACYCLINE

ABSTRACT

UDE SOJUNG. 2013. Determination of Withdrawal Time  of Oxytetracycline and its Effication On Tilapia (Oreochromis niloticus) infected by Bacteria Streptococcus iniae. Supervised by MUSTAHAL and DODI HERMAWAN

The purpose of this study was to determine the withdrawal time of ocxytetracycline (OTC) until no longer content of residues discovered in fish meat below the maximum residue limit (MRL). Also to determine the effectiveness of OTC medication in tilapia that was infected by sterptococcus iniae, through study withdrawal time (WT ) and drug effication test. Withdrawal time test was done by giving  75 mg OTC / kg body weight in tilapia (Oreochromis niloticus), mixed into the feed (orally) and administerd during 10 days for 48 day observation period. The residue of OTC was analyzed by using ELISA method. Drug effication test was carried out by using tilapia fish infected by bacteria S. iniae. The bacteria were injected intra-peritoneally (IP) about 0.1 ml at 10⁷ cells / ml,. Tilapia were fed with pellet at 75 mg / kg body weight for 10 days and observed  during 14 days. The results showed that the concentration of WT = 1 MRL (100 ppb) values ​​obtained on the day 5. the intersection of the linear regression line between the upper tolerance limit with 95% confidence limit of If the withdrawal time is determined by the concentration limit WT = 0.5 MRL (50 ppb), the time the drug was stopped 14 days after the last drug delivery applications. The observation parameter test fish survival rate of infected bacteria S. iniae showed a value of 60.00 ± 8.16%. Based on the results of analysis of variance the three treatments were (negative control, infected fish without medication and infected fish with medication) significantly different (P <0.05) in the decreasing the survival  rate of tilapia.
